STUDI IN-SILICO SENYAWA UMBI LOBAK PUTIH (Raphanus sativus L.) SEBAGAI KANDIDAT ANTI INSOMNIA

Insomnia adalah gangguan tidur yang umum dan tersebar luas yang disebabkan oleh berbagai faktor risiko.
Insomnia jangka panjang dapat menyebabkan masalah serius termasuk kecemasan, nyeri kronis, penyakit
kardiovaskular, hingga gagal jantung. Pengobatan insomnia konvensional memberikan kemanjuran moderat
tetapi dapat menyebabkan berbagai efek samping termasuk sedasi, penambahan berat badan, keluhan
gastrointestinal, insomnia rebound dan efek samping lainnya. Penggunaan bahan alam dipercaya memiliki
efek samping minimal sebagai pengobatan alternatif. Salah satu bahan alam yang dipercaya memiliki efek
sebagai anti insomnia adalah lobak (Raphanus sativus L.). Penelitian ini bertujuan untuk mengeksplorasi
senyawa dari umbi lobak (Raphanus sativus L.) sebagai kandidat obat anti insomnia. Dilakukan penelusuran
senyawa melalui studi docking in-silico menggunakan aplikasi AutoDock®. Hasil penambatan senyawa R.
sativus menunjukkan semua senyawa dapat berinteraksi dengan sisi aktif reseptor GABA. Interaksi terbaik
ditunjukkan oleh senyawa 13, 14, dan 16, dengan nilai energi bebas ikatan -9.73, -9.38, dan -7.74 kkal/mol,
sedangkan ligan alami diazepam memiliki nilai energi bebas ikatan -8.4 kkal/mol. Interaksi antar molekul
dengan residu asam amino yang paling mirip dengan ligan alami adalah Tyr58, Val203, Tyr210, Tyr160, Phe100,
Phe77, His102, Ser205. Dapat disimpulkan bahwa ketiga senyawa tersebut diprediksi berpotensi sebagai agen
anti insomnia.
