FORMULASI KAPSUL EKSTRAK DAUN KANGKUNG (Ipomoea aquatica Forsk) SEBAGAI KANDIDAT SEDATIVUM MENGGUNAKAN VARIASI KONSENTRASI POLYVINYLPYRROLIDONE

Kangkung mengandung senyawa flavonoid yang diketahui memiliki efek sedatif. Tujuan dari penelitian ini adalah untuk membuat kapsul dari ekstrak etanol kangkung (Ipomoea aquatica Forsk) dengan variasi konsentrasi polivinilpirolididon (PVP) yang stabil secara fisik. Ekstrak Kangkung diformulasikan menggunakan PVP sebagai pengikat, dengan konsetrasi 5%, 10% dan 15%. Setiap formula dievaluasi stabilitasnya menggunakan climatic chamber. Hasil uji stabilitas menunjukkan formula yang mengandung PVP 5% stabil secara fisik. Efektivitas kapsul sebagai sedativum kemudian dievaluasi menggunakan mencit jantan (Mus musculus L.). Hewan dikelompokkan menjadi empat kelompok, diberi Natrium CMC, diazepam 2 mg / kg BB, kapsul kangkung 40 mg / 20 g BB dan 80 mg / 20 g BB. Empat puluh lima menit setelah diberi sediaan uji, hewan diberi sodium thiopental i.p. Kemudian onset dan durasi diamati. Hasil penelitian menunjukkan serbuk kapsul yang mengandung ekstrak kangkung (Ipomoea aqutica Forsk) memiliki aktivitas memperpanjang durasi tidur pada mencit. Analisis statistik menggunakan ANOVA menunjukkan ada perbedaan waktu durasi yang signifikan antara kelompok yang diberi natrium CMC dan kelompok yang diberi serbuk kangkung dosis 40 mg/20g BB dan 80 mg /20g BB. Hal ini menunjukkan adanya efek sedatif kapsul yang dibuat dari ekstrak kangkung.
