KARAKTERISASI SENYAWA DAN UJI AKTIVITAS ANTIBAKTERI DARI ISOLAT Actinomycetes rhizosfer TANAMAN TEH (Camellia sinensis L.)

Actinomycetes merupakan genus yang paling dominan dalam kelompok bakteri penghasil antibiotik dengan 80% dari turunan antibiotiknya telah diidentifikasi. Tujuan dari penelitian ini adalah untuk menentukan karakter senyawa metabolit sekunder dan aktivitas antibakteri dari senyawa isolat Actinomycetes rhizosfer Camellia sinensis L. Diperoleh tiga isolat dari hasil isolasi dan diberi nama berturut-turut TH.1, TH.2 dan TH.3. Skrining aktivitas antibakteri dilakukan dengan uji antagonis. Isolat dengan aktivitas antibakteri tertinggi dilanjutkan untuk tahap selanjutnya. Dilakukan fermentasi selama 14 hari untuk memperoleh metabolit sekunder dari isolat Actinomycetes. Hasil penelitian menunjukkan bahwa ekstrak etil asetat dari isolat Actinomycetes memiliki aktivitas antibakteri terhadap bakteri Staphylococcus aureus ATCC 25922 dan Escherichia coli ATCC 25922. Pada uji Kromatografi Lapis Tipis diperoleh hasil positif senyawa golongan alkaloid. Berdasarkan pengujian karakterisasi isolat dengan Spektroskopi UV-Vis dan FTIR diperoleh hasil senyawa isolat Actinomycetes merupakan senyawa alkaloid dengan panjang gelombang 271.00 nm dengan nilai absorbansi 0.727 dan mempunyai gugus fungsi N-H pada serapan 2852.81 cm-1 dan pada serapan 2922.25 cm-1 yang merupakan ciri khas dari alkaloid.
